This kit introduces the use of restriction enzymes as tools to digest DNA at specific nucleotide sequences. Bacteriophage Lambda DNA has a linear structure and 5 Eco RI recognition sites. Separation by agarose gel electrophoresis of an Eco RI digest of lambda DNA will yield 6 bands (5 distinct bands; 2 are very close in size) corresponding to the DNA fragments.
REQUIREMENTS: Horizontal gel electrophoresis apparatus; D.C. power supply; visualization system (white light); automatic micropipet with tips; pipet pumps or bulbs; 250ml flasks; hot gloves and safety goggles; metric rulers; distilled or deionized water; waterbath; microwave oven or hot plate; photodocumentation system (optional); balance; latex or vinyl gloves; large weigh boats for destaining.
